background: Integrins are heterodimers composed of noncovalently associated transmembrane alpha and beta subunits, which heterodimerize to produce more than 20 different receptors. Most Integrin receptors bind ligands that are components of the extracellular matrix, including Fibronectin, collagen (COL) and Vitronectin. Certain Integrins can also bind to soluble ligands, such as Fibrinogen, or to counterreceptors on adjacent cells, such as the intracellular adhesion molecules (ICAMs), leading to cellular aggregation. Integrin ?1, also known as ITGA11 or MSTP018, is a 1,189 amino acid single-pass type I membrane protein that contains one VWFA domain and seven FG-GAP repeats. Expressed at high levels in heart and uterus and present at lower levels in pancreas, kidney, skeletal muscle, placenta, lung, colon and brain, Integrin alpha 11 exists as a heterodimer with Integrin beta1 and functions as a receptor for collagen. Function: Integrin alpha-11/beta-1 is a receptor for collagen. Subcellular Location: Membrane. Tissue Specificity: According to PubMed:10464311, highest levels of expression in uterus and heart, intermediate levels in skeletal muscle and intermediate to low levels in pancreas, kidney and placenta. According to PubMed:10486209, also found in brain, colon, lung, small intestine, stomach, testis, salivary glands, thyroid glands and prostate. Very low levels in peripheral blood lymphocytes, fetal brain and fetal liver. Similarity: Belongs to the integrin alpha chain family. Contains 7 FG-GAP repeats. Contains 1 VWFA domain. Protocol
The cells were fixed with 4% PFA (10min at room temperature)and then permeabilized with 90% ice-cold methanol for 20 min at -20℃. The cells were then incubated in 5%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. Acquisition of 20,000 events was performed.
